OPTIMISATION OF CULTURE CONDITION FOR SACHA INCHI (Plukenetia Volubilis) CALLUS INDUCTION

Plukenetia volubilis or commonly known as sacha inchi is reported to produce wide range of health-promoting bioactive metabolites. These metabolites functions supplements in eradicating various types diseases. Sacha has large edible seeds that are rich phenolic content, minerals and essential fatty acid, such omega 3 (ω-3), 6 (ω-6), 7 (ω-7), 9 (ω-9). In vitro cultures could serve alternative producing many compounds. As an initial step towards initiating culture, the effect 2,4-D TDZ on inducing callus were investigated. Two different explants, leaf male flower used this study. Surface sterilization was first optimized overcome culture contamination. The most effective surface by using 70% ethanol (30 seconds) 0.5% sodium hypochlorite (8 minutes), which resulted 82.5% 95% survival rate for explants respectively. Next, calli induction cultured MS medium supplemented with concentrations TDZ, either alone combination grown at 24 hours dark photoperiods. morphology size observed. results obtained from experiment varied depending treatments, friable compact creamy white, pure white brownish colour. For both, 3% (w/v) sucrose 1.0 mg/L 0.005 recorded best response term size, forming callus.
